Upper Organic Phase

what is organic phase?

i have a mixture of polymer (PHB) in Methonol+KOH which was incubated for 1 hour at 100C and then added chloroform and dist water. the organic phase has to be injected into GC? which is oganic phase… the upper pahse or lower phase. Please help me.. Urgent!!

Since you have used chloroform the organic phase is the lower phase. Chloroform is more dense than water.

If you are unsure take the whole mixture in a separating funnel. Draw a line on the glass where the phase boundary is seen. Add a little more organic solvent. If the phase boundary is now above the drawn line then you know that the organic solvent is the lower phase. If the phase boundary is still at the drawn line then you know that the organic phase is the upper phase.
